Repair of injured mammlian central nervous system (CNS) is one of the most interesting fields in neuroscience research .It is being a puzzling problem in neuroscientists. In recent years, with the development of Neural stem cells (NSCs) research, people have further realization of the nervous system's growth process, treatment of inducing the cell divide, therapy of spinal cord hurt. The present study is about the in vitro cell culture, proliferation and differentiation of the neural stem cells (NSCs), especially the spinal cord derived neural stem cells which provide new evidence to the clinical application of the spinal NSCs.Our study is to look for a seed cell for repairing of spinal cord injured. There are two parts in the study. In the first part, we isolated clone cells from E14 fetus mice's spinal and striatum through non-serum cell culture. Immunohistochemistry was used to identify the cell type .All the data were analyzed by statistic. The results are as follows: (1)The spinal cord derived NSC and the striatum derived NSC all express Nestin antibody.(2) After these NSC differentiation, the mature cell differentiated into neurons, astrocytes and oligodendrocytes. From this part of experiments, we can draw a conclusion that the multipotental NSC can be isolated from spinal cord and striatum of mice, but there are differences in culture situations and methods.In the second part of this study, the objective is to investigate the effects of BMP-2 on the inducible differentiation of spinal cord derived NSCs into neural...
