Objective: To study the changes and their significances of Granule Membrand Protein 140 ( GMPi40 ) > thromboxane B2 ( TXB2 ) , 6-Keto-prostaglandin-1 (6-keto-PGF i a ) plasma levels of dog models with carotid atherosclerosis.Methods: The dog atherosclerosis models were made by balloon catheter denudation of the dog carotid artery and then two months feeding of animals with food rich with fat. Twelve dog models entered this study and their basic (before balloon injury) GMPi40> TXB2 and 6-keto-PGFia plasma concentrations were used as control. Following, 4ml peripheral venous blood from each dog was taken as series of samples within 24 hours and after the procedure every other week for eight weeks in all. The plasma levels of those markers were measured with ELISA and compared with those of control, respectively. Data are presented as mean 盨D and paired students t test was applied for comparison between their basic values and those after the procedure a week apart. The level of P value less than 0.05 was considered significant.Results : The GMPi4o. TXB2. 6-keto-PGF1. plasma levels of dog models within 24 hours after balloon injury are obviously higher than those of control, for GMP140 (ng/ml) being 18.046?.122 vs 9.171?.122,TXB2(pg/ml) being 132.053?1.124 vs 34.572 ?8.031 and 6-keto-PGFla(pgdnl) being 120.224 vs 30.025?5.212 (PO.001). And each of GMPuo and TXB2 plasma levels of the models were higher than that of control (p<0.05). Whereas, the plasma levels of 6-keto-PGFia seemed to be no difference between the control and the samples after the model made.Conclusion: Our dog models had almost all pathogenical features of atherosclerosis, which would be useful in various studies about the diseases.These results indicated that balloon injury to dogs carotid artery enhanced platelet activities, its aggregation and endothelial cells activities. The results demonstrated that the formation of animal models' carotid atherosclerosis accompliced with provoked platelet activities at least for eight weeks after the injury.It suggested that the interaction between platelets and leukocytes appeared to be the key to the inflammatory response during the formation of atherosclerosis. So, by understanding of activation and aggregation of platelets to evaluate the risks of arterial restenosis and embolization after percutaneous transluminal angioplasty is beneficial to the patients with atherosclerosis.
