Objective To study the expression of a novel inhibitor gene of apoptosis survivin in cervical carcinoma and its association with the expression of HPV16-E6 and p53 genes.Materials and Methods A total of 116 uterin cervicle tissue samples including 72 cases of squamous cervical carcinoma ( SCC ) tissues, 23 cases of CIN tissues and 21 cases of normal cervical tissues were examined in this study.The SCC specimens consisted of 31 cases of stage â…  , 28 cases of stage â…¡ , 13cases of stageâ…¢.The mean age of the patients was 53.6 years, (SD, 11.84 years; range, 27 ~ 71 years ) .The CIN specimens consisted of 6 cases of CIN â…  , 9 cases of CIN â…¡ , 8 cases of CIN â…¢. The mean age of patients was 51.9 years, (SD, 10.62 years; range, 29 ~ 67years ).The mean age of normal cervical women was 52.4 years, ( SD, 11.48 years; range, 31~65years) .Using streptavidin-biotin peroxidase(SP) method of the immunohistochemistry,we examined the expression of the proteins survivin , p53 and HPV16-E6 in normal cervical tissues,CIN and SCC.The expression of survivin in different sample were not same. It was associated with FIGO stageand historical grade.Statistical analysis was performed by using SPSS11.0 statistics software.Results 1.The protein of survivin was expressed in 52 of 72(72.2%) cases of squamous cervical carcimomas and in 4 of 23 (17.39% ) cases of CIN,in contrast,no expression was found in normal cervical tissues.Among SCC group, The expression of survivin protein in low-stage (Stage â…  ) and low-grade(G1. G2)were significantly lower than that in high-stage (Stage II and III )and high-grade(G3)( p< 0.05,p< 0.01).It was associated with lymphatic metastasis (p < 0.01)but it has no association with the age of patients(p < 0.05).The expression of survivin protein was related to FIGO stage and historical grade. (p<0.05).2.Survivin protein was expressed in 81.39% ( 35/43 ) of HPV16-E6 positive cases.However, it was expressed in 58.6%( 17/29 ) of HPV16-E6 negative cases. The expression of survivin protein was strongly associated with the expression of HPV16-E6(p< 0.05).3.Survivin was expressed in 84.78%(39/46) of p53 positive cases however it was expressed in 50%( 13/26) of p53 negative cases.The expression of survivin protein was strongly associated with the expression of p53 (p < 0.01).Conclusions 1.Survivin protein reexpressed and overexpressed in squamous cervical carcinoma tissues.
