Experimental Study Of EGb761 Treatment For Hypertensive Intracerebral Hemorrhage In Rats

Objectives: To investigate the expression of nuclear factor-kappa B(NF- k. B) and the change of brain water contents in focal brain tissue around hematoma after hypertensive intracerebral hemorrhage (HICH) in rats; to discuss the protective effect of treatment with Extract of Ginkgo biloba 761 (EGb 761) for HICH.Methods: 96 adult female Wistar rats were randomly divided into normal control group (n=24), false operation group (n=24), HICH model control group (n=24) and EGb 761 treatment group (n=24). In HICH model rats, the anterior branches of bilateral renal arteries were coagulated partly, then the animals were raised for 6 weeks with high salt, fat and cholesterol food, then intracerebral hemorrhage was induced by injection of autologous blood into the right caudate nucleus. In false operation group, nothing was injected into normal rats' caudate nucleus, and in normal control group the animals were normal rats. EGb 761 treatment was given at 30 minutes after HICH in EGb 761 treatment group. Everyday behavioral function was evaluated once before and after HICH from day 1 to day 7. Brain water content was measured by the dry-wet method at 48 hours after HICH. Immunohistochemical technique was used to evaluate NF- k B expression in focal brain tissue around hematoma at 72 hours after HICH.Results: Neurobehavioral function test of EGb 761 treatment group was better than HICH model control group after day 3, showing significant difference (t=3. 564-4. 320, P<0. 01). The test of normal control group and false operation group were better than the other group, showing significant difference (F=478. 55-2388. 32, q=33. 085-89. 833, P<0. 01). Compared the brain water content in hemorrhagic hemisphere, HICH model control group was worse than EGb 761 treatment group, showing significant difference (t=10. 546, P<0. 01). There was significant difference between hemorrhagic hemisphere and non-hemorrhagic hemi- sphere in both two groups (t=20. 457, 14. 563; P<0. 01). In hemorrhagic hemisphere, the NF- k. B cells of EGb 761 treatment group expressed markedly less than the ones of HICH model control group, showing significant difference (t=3. 419, P<0. 01). In both EGb 761 treatment group and HICH model control group, the NF- k B cells of hemorrhagic hemi- sphere expressed markedly more than the ones of non-hemorrhagic hemisphere, showing significant difference (t=7. 279, 6. 952,'P<0. 01).
