The Relation Between Expression Of Il-8, Icam-1 In Tissue Surrounding The Hematoma And Brain Edema In Experimental Intracerebral Hemorrhage And The Intervention Role Of Injection Of Ginkgo Biloba Leave Extract

Objective To investigate the relationship between expression of interleukin-8 (IL-8)and intercellular adhesion molecule-1 (ICAM-1) in tissue surrounding the hematoma in rats after intracerebral hemorrhage (ICH) and brain edema, approach the protection of injection of ginkgo biloba leave extract (Ginaton) on inflammatory reaction and provide experimental basis for treatment of ICH. Methods The ICH model in rats of this study was established by heparinizedⅦ-type collagenase into left caudate nucleus, and the effects of Ginaton on it were observed. 160 rats were randomly divided into five group: sham operated group,ICH group and Ginaton high,middle,low three dose group. Every group was divided into 6 h,24 h,72 h,7 d four time points. Every time points have eight rats. Berderson score to evaluate behavioral change at every time point, the dry/wet weight method to check up cerebral water content, pathological change were collected for observation under lightmicroscop, the expression of IL-8,ICAM-1 were detected by immunohistochemistry. Results The effect of Ginaton on the score of nerve function after ICH in rats: The score of nerve function was obviously increased after ICH in rats, emerge obvious neurologic impairment, 24 h to achieve peak (P<0.01). The Ginaton high,middle dose group can decrease the score of nerve function in rats (P<0.05 or P<0.01). The Ginaton low dose group also depress the score of nerve function, but no significant difference compare with ICH group (P﹥0.05); The effect of Ginaton on the brain edema after ICH in rats: The content of edema in rats 24 h begin to heighten after ICH, were highest at 3 d (P<0.05 or P<0.01), reached the normal levels at 7 d. The Ginaton high,middle dose group 24 h,3 d obviously statistic significance with the Ginaton low dose group and sham operated group (P<0.05). The Ginaton low dose group can depress the content of edema, but all no significant difference compare with ICH group; The effect of Ginaton on pathological change after ICH in rats: Tissue surrounding the hematoma have obviously inflammatory reaction after ICH in rats, have small amounts inflammatory cell infiltrate at 6 h, begin to heighten about neutrophil inflammatory cell infiltrate at 24 h, hematoma environ and within hematoma gliacyte,blood vessel hyperplasia at 3 d, hematoma obviously deflate, gliocyte and obviously blood vessel hyperplasia at 7 d. Ginaton treated group can make inflammatory reaction soften, the hematoma obviously deflate, a great quantity blood capillary hyperplasia; The effect of Ginaton on IL-8 after ICH in rats: In sham operated group can see small amounts IL-8 masccline cell at tissue surrounding pin hole, the main cell is minute vascular endothelial cell; In ICH group begin to heighten at 6 h, the main cell is vascular endothelial cell,gliocyte,neuron, were highest at 24 h (P<0.01), were decreased at 3 d, still have small masccline cell at 7 d. Ginaton can make IL-8 decreased, the higher dose of the more obvious effect; The effect of Ginaton on ICAM-1 after ICH in rat: In sham operated group can see small amounts ICAM-1 masccline staining blood vessel in tissue surrounding pin hole, no neuron express; In ICH group tissue surrounding the hematoma have ICAM-1 positive reaction blood vessel and nerves cell, obviously statistic significance with sham operated group (P<0. 01), 6 h begin to heighten, were highest at 72 h (P<0.01), still have small masccline cell at 7 d. Ginaton treated group ICAM-1 decreased significant, the higher dose of the more obvious effect; The relative relationship between the changes of IL-8 and ICAM-1 content and brain edema: The summit of IL-8 levels and the summit of ICAM-1 levels coincide with the summit of brain edema. Conclusion There were inflammatory cell infiltration,inflammatory factor express conducted by IL-8,ICAM-1 which led to damage after ICH, and have time associativity with brain edema after ICH. Prompted the two may be related with inflammatory reaction after ICH; The IL-8 levels and the ICAM-1 levels coincide with the summit of brain edema. Prompted both may be related with the occurrence and development of brain edema; Ginaton can significantly decrease neurological scale, reduce brain edema, improve pathological change, decrease the levels of IL-8 and ICAM-1 after ICH, intercept inflammatory reaction. It can provide effective help for treating the cerebral injury. The protective effection of Ginaton was dose dependent.
