Objective: To study the effect and mechanisms of arsenic trioxide on SGC7901/ADM gastric cancer cell line. Methods: AS2O3 was used on SGC7901/ADM and SGC7901/S gastric cancer cell line with different concentration and in different time (l.The growth of the cell line was studied using MTT assay.(1)To determine the sensitivity of the SGC7901/ADM and SGC7901/S cell line to ADM.(2) To determine the sensitivity of the two group to AS2O3.(3) To determine the sensitivity of the two group to ADM after the two groups were interfered by AS2O3). 2.Cell cycle and apoptosis rate were analysed by the flow cytometiy.3.To analyse the express of caspase3 in SGC7901/ADM interfered by AS2O3 with immunohistochemistry .4.T0 determined the express of caspaseS in SGC7901/ADM interfered by AS2O3 with Western Blot. Results: l.The distribution of the two group cell cycles changes after interfered by AS2O3 with different doses. Ratio of G0-G1 cells enhanced following concentration and time of AS2O3. The ratio of G0-G1 cells of no-drug groups and CsA groups was steady, not vary with time. 2.Cell apoptosis rate rose with the dose of AS2O3, there was statistical difference with every dose group in 24hrs groups ;there was difference between 24hrs and 48hrs 72hrs groups in the difference time in the same dose of AS2O3. There was no difference between 48hrs and 72bxs in the highest dose. 3. The positive grains of caspaseS primarily located cytoplasm near the nucleumembrane. The express of caspase3 in SGC7901/ADM cell line was little without the interference of AS2O3, the brown grains were little and the color was light. The brown grains gradually increased namely the express of caspaseS increases with the increase of dose and time. Conclusion: 1.SGC7901/ADM cell line is resistant to ADM. 2.SGC7901/ADM and SDC7901/S have the same sensitivity to As2O3. 3.AS2O3 can reverse the resistance to ADM of SGC7901/ADM, the more of the As2O3, the longer of the function time, the more powerful of the reversing MDR. 4.The function of reversing MDR may be related to caspaseS protein family of apoptosis roads.
