Tumor Suppressor Gene PTEN,P53 And The Dualistic Model Of Endometrial Carcinoma

Objective: Endometrial carcinoma is one of the most common femalepelvic malignancy.In those years ,scientists never stop researching itsmechanism on molecule level.However ,there is no agreement among manyviews.Scholar Lax put forward the new standpoint of dualistic model forendometrial carcinoma in 1997 — — type Ⅰ is estrogen –dependentcarcinoma,endometrial endometrioid carcinoma (EEC);type Ⅱ is estrogen–independent carcinoma,including serous carcinoma and clear cellcarcinoma .The experiment will assay the difference of PTEN P53protein expression between two types of carcinomas and make sure that thedifferent mechanism of tumors occur in two types of tumor. Methods: There is 20 cases typeⅠtumors including 15 cases tumors instage Ⅰ,1 cases stageⅠa-Ⅰb,4 cases stageⅠc and 5 cases in stageⅡ,Ⅲ andⅣ;5 cases in the group of lymph node metastasis , 15 cases in the group ofnon-lymph node metastasis.There is also 15 cases type Ⅱ tumors consist of 8cases in stageⅠ-Ⅱ and 7 cases in stage Ⅲ-Ⅳ between 5 cases in the group oflymph node metastasis. 10 cases in the group of non-lymph nodemetastasis.The study will show the relationship between PTEN,P53 proteinexpression and two types of carcinoma ,the study will also identify thesignificance of two tumor suppressor genes in different clinical grades ,pathological grades and lymph node metastasis by immunohistpchemistry. Results: 1.Of two types of tumors , PTEN protein lack expression mainly intype Ⅰ.There is significantly difference in two types. (P<0.01) 2.PTEN protein lackness expression correlate well with clinical grades,more earlier stage more higher lackness protein expression.(P<0.05),notcorrelate with pathological grades and lymph node metastasis. (P>0.05) 3. P53 protein express mainly in type Ⅱof two types of tumors.It issignificant. (P<0.01) 4. P53 protein expression correlate positively with clinical grades andpathological grades, not correlate with lymph node metastasis. (P>0.05) 5. P53 protein express highre in serous carcinoma than in clear cellcarcinoma . It is significant. (P<0.05) 6.The difference of P53 protein expression in PTEN lack expressionand P53 protein lack expression in PTEN protein expression are significant.(P<0.05) Coclusion: 1.The sudden change of PTEN happpen in type Ⅰtumors easily indualistic model of endometrial carcinoma;PTEN protein express in earlyclinical grade mostly to show that it is a early molecule case. 2. The sudden change of P53 happpen in type Ⅱtumors mainly indualistic model of endometrial carcinoma;P53 protein expression occur inlately in clinical grade mostly to show that it is a later peroid molecule case.And in type Ⅱtumor mechanism research work on molecule level ,theprobability of sudden change of P53 is higher in serous carcinoma than inclear cell carcinoma. 3.There is a kind of concern harmony relationship between PTEN andP53 in the happeness and development of endometrial carcinoma.
