Objective: to investigate the roles of semen in regulation of endometrial stroma cells decidualization and embryo development Methods:1.Stroma cells derived from human endometrium during the late proliferate phase or secretive phase were treated with estradiol,progesterone to induce decidualization. Spermatoza and seminal plasma, isolated from normal men were added to to the stroma cells. The stroma cells morphologic changes and prolactin (PRL)production in the media were examined and compared.2. The 2-cell mouse embryo were added to the stroma cells after cultured 8 days. The embryo growth rate and blastocyst formation rate,hatching,adhereing,outspreading of mouse embryo and the development of stroma cells were observed under the microscope. 3. In order to observe the effect of spermatozoa on implantation rate,the blastocysts were transferred to day 3 fake pregnant female mice and control group . Results: 1. Seminal plasma (SP)significantly increased the degree of the stroma cells decidualization compared with spermatoza and the control group.PRL secretion of co-cultured with SP was significantly higher than the other two groups(p<0.05). 2.SP significantly promoted the embryo growth rate (after 8-cell)and blastocyst formation rate,hatching,adhering,outspreading on the endometrial stroma cells(p<0.05). 3. The implantation rate of two groups were no difference , spermatozoa have no effect . conclusion:1.SP can contribute to the stroma cells decidualization and improve PRL secretion.2.SP can promote the embryo development in co-cultured with endometrial stroma cells. .3. The implantation rate between ovi-duct ligated group and pseudopregnancy group was no difference ,indicate the effect of seminal plasma and semen on blastocyst implantation may be similiar .
