2-Methoxyestradiol(2-ME), an physiological metabolote of estrogen, is a promising anti-tumor drug.It has been shown that 2-ME induced a series of Ewing's sarcoma cells, such as EW7, SK-N-MC to apoptosis. Nuclear factor κ B( NF-κ B) is thought to be involved in many apoptotic induction by various signal. To investigate the effect of 2-methoxyestradiol (2-ME) on NF- κ B activation and its role in apoptotic induction during 2-ME -induced apoptosis of SK-N-MC Ewing's sarcoma cells, this study is divided to 2 parts:Part 1: The change of NF- κ B activity during 2-ME -induced apoptosis of SK-N-MC Ewing's sarcoma cellsAim: To investigate the change of NF- κ B activity during 2-ME -induced apoptosis of SK-N-MC Ewing's sarcoma cells.Methods: To assess the apoptotic cells induced by 2-ME in SK-N-MC cells with Annexin V/PI flow cytometry.
