Objective To study the protective effects of ulinastatin on mitochondrial function during ischemic model of total hepatic I/R injury in dog. Methods The hepatic function and the activity of H~+-ATPase were determined. The ultrastructures of mitochondria were observed by electron microscope. Results The hepatic function was improved in experiment group than in control group .The activity of H+-ATPase was 1.78±0.07 in experiment group and 1.50±0.08 in control group after 15 minutes of ischemia(p<0.05).It was 1.47±0.10 in experiment group and 1.09±0.07 in control group after 60 minutes of ischemia(p<0.05).The damage of mitochondrial ultrastructure in experiment group was significant lower compared with control group. Conclusions Ulinastatin has protective effects on mitochondrial function of hepatic ischemia-refusion injury. The theory is to stablize mitochondrial membrane structure and to improve activity of H~+-ATPase.
