Purpose: Genetic predisposition is proven related with onset of periodontitis. Experts in genetics have found genetic factors contribute a lot to the onset and development of periodontitis after analyzing it by way of kindred analysis and Mendel's law. Presently, a lot of scholars have studied it genetically and even achieved great progress: Coded genetic polymorphism of inflammatory media may directly regulate the inflammatory reaction of host to bacteria and coded polymorphism of interleukin IL, tumor necrosis factor a, and Ⅱa, Ⅲb of Fcγ receptor may form predisposing factors of periodontitis.In our previous studies, we found that CGRP exists extensively in periodontal tissues fibers, especially at the beginning foci of periodontitis. But no report on the relation between CGRP gene polymorphism and periodontitis is delivered up to now. In order to further probe into the relationship between them, we have studied the relation between 4 CGRP-coded gene polymorphism sites and severe adult periodontitis. Methods: 1.Cases and grouping: All the subjects were from Guangdong ProvincialStomatology Hospital: Patients with severe adulty periodontitis asexperimental group and health adults as controls.2.Chelex-100 and Chris-Hcl were adopted to extract DNA from buccal mucosa swabs. followed by purifying the PCR products. 3.The gene sequencing was conducted on the target gene to identify thesite of CGRP gene polymorphism.Results:In comparison of the two methods for extracting DNA, Tris-HCl was found to be simpler, though they showed no significant differences in DNA concentrations and quality. The sequencing was analyzed by BLAST, showing that the amplified sequence of CGRP was to a great extent homogenic to that by NCBI (99%) and that the site of CGRP+5247 demonstrated polymorphism of mononucleotide,significant difference by two group.(x~2=15.14,p<0.001 ).
