As it is all known that,growth and metastasis of tumor depend on the forming of newborn blood vessel, and tumor cells can secrete vascular endothelial growth factor(VEGF) and its receptor which accelerate the forming of newborn blood vessel. Therefore,it is of great importance to study the expression regulation of VEGF to elucidate the mechanism of cancer metastasisSTK11/LKB1 (serine theronine protein kinase) is a tumor supressor gene newly identified in patients affected with Peutz-Jeghers syndrome.Till now,a large amount of researches related to tumor suppress gene have been conducted.For example,studies shows that tumor suppress gene p53 or PTEN can suppress the expression of VEGF in tumor cells. And pathological study demonstrated that mutation of LKBl contributes to vascular abnormalities in embryo and up-regulation of VEGF in embryonic fibroblast cells. Also, in 58 percent of those patients with lung carcinoma metastases to the brain, mutation of LKBl gene happened.lt can be infered from these materials that LKBl may play a role in the synthesis of VEGF in tumor cells.In order to elucidate the role of LKBl in controlling bionomics of tumor.the following studies have been done.First of all,the status of LKBl gene expression in A549 cell lines of human lung adenocarcinoma which inclinate to metastasis and in 95D or 95C cell lines of human megacell lung cancer which do not inclinate to metastasis.Then,the role of LKBl in controlling the synthesis of VEGF in a model of acquired expression of LKBl was studied.Finally,the role of LKBl in controlling the synthesis of VEGF in a model of acquired silencing of LKBl expression was studied.The aim of this research is to elucidate the role of LKBl in controlling the synthesis of VEGF in lung cancer cells.First.lung cancer cells A549,95C and 95D culture in vitro were studied by Northern Blot...
