Lung cancer is a severe disease that has a great danger impact on human health. Despite great improvement of medicial treatment, there is no more than 10% livability when getting a lung cancer. The biological character of lung cancer is the metastasis, which is also the first reason of the treat failure and high mortality. The angiogenesis is the basis of the tumor growth and metastasis, how to inhibit the angiogenesis is the important method in the treatment of cancer. As a cytokine, vascular endothelial growth factor (VEGF), that is an important role on the vascular growth. Many research revealed that VEGF is a key factor to affect the vascular growth and the metastasis for the lung cancer. So the inhibition of the VEGF expression will be helpful to arrest the growth and metastasis of the lung cancer. So we select the gene of VEGF as the interest gene and try to find an effective method for tumor therapy by PCR-based siRNA expression cassettes to inhibit VEGF expression in non-small cell lung cell A549.Gene carrier is an important part in the gene therapy. There is some defect for viral and non-viral carrier used in recent treatment of gene therapy. In our experiment, we select a non-viral carrier from a Japanese company NDT for our future experiment. We had got good transfection efficiency up to 40% by improving the transfection condition. Then we designed the siRNA sequence targeting the CDS region of VEGF, then utilized PCR based strategy for rapid synthesis of siRNA expression cassettes.
