Lung cancer is an epithelial cancer arising from the bronchial surface epithelium or bronchial mucous glands. Non small cell lung cancer constitutes about 75%~80% of all lung cancer.It is one of the leading causes of cancer death in mankind with five—year survival rate remains 15 %. At the time of diagnosis, a lot of people have got unresectable stage non small cell lung cancer. Both chemotherapy and radiotherapy are widely used in unresectable stage non small cell lung cancer.But non small cell lung cancer cells have both low radiosensitivity and chemosensitivity. How to enhance the radiosensitivity or chemosensitivity of non small cell lung cancer cells is a key point of oncology and radiation biology which has great value and meaning.Signal transducers and activators of transcription 3 (STAT3) pathway can be activated by cytokines and growth factors, and activation of STAT3 is involved in modulating cell proliferation, differentiation and apoptosis. STAT3 has been classified as an oncogene because STAT3 can mediate malignant transformation of cultured cells.Many studies show that there was obvious difference of STAT3 expression between the poor differentiated lung cancers and the well differentiated lung cancers. The expression of STAT3 was higher in lymph node metastatic cancers than in non lymph node metastatic cancers. No obvious difference of STAT3 expression was found in different tumor size or tumor type. The expression of STAT3 may contribute to malignant transformation of non small cell lung cancer.Antisense technology is a method toinhibit, block, or destroy target genes by use of antisense DNA, antisense RNA , and ribozyme etc. The process in replication, transcription, or translation was blocked by specific,complementary, and pairing combination of antisense molecule with target genes.
