Objects: To observe whether antisense-EGFR could improve the sensitization to the radiation in lung adinocarcinoma cell line and to offer the found for radiotherapy combined with targeted therapy in the clinical work.Methods: We examined the expression level of EGFR in spc-a-1 cell lines with immunohistochemical. The spc-a-1 cell line was transfected with antisense-EGFR-pcDNA3 by lipofectium 2000 and control group was transfected with pcDNA3. Select cell clones stable expressing antisense-EGFR using G418 and detect the suppression of the expression of EGFR mRNA and protein by RT-PCR and Western Blot experiments, testify the influence of antisense-EGFR on proliferation of cells by growth curve and cell cycle assay. Then we gave them radiation 800cGy with 6MVX , observing apoptosis after radiation 48 hours and cell growth curve . Results: Our results showed that the cells cycle arrested in the G2, M stage in the antisense-EGFR group was higher than the other tow groups (p<0.05). The growth of the antisense-EGFR group was delayed(p <0.05), but not observed in the other tow groups. We found that the apoptosis of spc-a-1 cells in the antisense-EGFR+radiation group was obviously higher and the growth was more inhibited obviously than the other four groups(p<0.01).Conclusions: The research demonstrated that antisense-EGFR could suppress the growth of cancer cell, improve the sensitizes of the radiation and provide the found for the next vivo experiment.
