Object: To study the effects of Omeprazole on the ROS ,mitochondrial membrane potential and expression of apoptosis –relatedgenes in SGC-7901 cell line. Method: In presence of Omeprazole, the proliferation and apoptosisratio of SGC-7901 cell were detected with MTT colorimetric assay andTUNEL. The expression of bax,bcl-2 and caspase-3 protein wereassessed by immunohistochemical assay. Flow cytometer was used todetermine the cell cycle,mitochondrial membrane potential and ROS. Results: Omeprazole significantly inhibited SGC-7901 cell lineproliferation, as well as increased the apoptosis ratio of SGC-7901 indose-dependent and time-dependent manner.Compared with control group,the expression of caspase-3 ,ROS were elevated, mitochondrial membranepotential was dropped. Significant alteration of the decreasing percentageof G1 phase and the increasing percentage of G2 phase of SGC-7901 cellline could be observed after incubating 24h with Omeprazole. There was noapparent change in bax and bcl-2. Conclusion: Omeprazole could significantly inhibited theproliferation and induced apoptosis of SGC-7901 cell by increasingintracellular ROS.
