Objective: To study the effect of reactive oxygen species (ROS) and Mitochondrial membrane potenial on {2-[(3- carboxy-1-oxoprogyl) amino]-2-deoxy-D-Glucose} (COPADG) inducing apoptosis in esophageal cancer cell line Eca-109.Methods: Eca-109 cells were cultured in RPMI-1640 medium. COPADG worked on Eca-109 cells for 24 hat different concentration (0.00 mmol.L-1, 0.02 mmol.L-1, 0.03 mmol.L-1, 0.04 mmol.L-1) . Cell growth inhibitory rate was determined by MTT assay. Then determine the apoptosis by flow cytometry with AnnexinV and PI at different concentration (0.00 mmol.L-1, 0.02 mmol.L-1, 0.03 mmol.-1, 0.04 mmol.L-1) . Dihydrogen rhodamine 123 (DHR123) as a ROS capture was incubated while the cells in the absence of COPADG. The fluorecent intensity of rhodamine 123 (Rh 123) the product of cell cellular oxidation of DHR123 was assayed by flowcymetry. The ROS levels was thus detected.△(?)m was determined by flow cymetry with Rhl23.Results: After the drug worked on the cells for 24 hthe cell growth inhibitory rate is 0.002±0.007, 0.726±0.006, 0.761±0.007, 0.822±0.008. At the concentration ranging between 0.00 mmol.L-10.04 mmol.L-1 the COPADG inhibited the growth of Eca-109 cells in a dose dependent manner. The apoptosis rate is 3.38 %, 52.6 %, 63.7 %, 79.8 % and the level of ROS is 62.3, 84.2, 95.1, 107.6. The correlation coefficient between ROS and apoptosis rate is 1.0 (p<0.01) . The Mitochondrial membrane potenial is 705.3, 662, 309.9, 280.The correlation coefficient between Mitochondrial membrane potenial and apoptosis rate is 1.0 (P =0.000). Conclusion: COPADG can induce esophageal cancer cell line Eca-109 cells apoptosis through evelating cellular ROS, COPADG can induce esophageal caner cell line Eca-109 cells apoptosis and redudce the Mitochondrial membrane potenial in a dose dependent manner.
