Objective: To observe the changes of γδT cells in blood of rats with osteosarcoma after treated with high intensity focused ultrasound(HIFU), and to investigate the relationship between the changes of γδT cells and the increased expression of heat shock proteins. Methods: After identifying the characteristics of UMR106 osteosarcoma in nude mice, transplanted the tumor tissue fragments orthotopically into the right tibias of rats. 3 weeks later, 4 normal rats were used as normal control named group1, 28 rats with osteosarcoma were divided into 4 groups named group 2, 3, 4 and 5 respectively. Rats in group 2 with no treatment, rats in group 3 were treated with HIFU, rats in group 4 were treated with quercitin(QCT) before HIFU, and rats in group 5 were treated with QCT only. At the day 1 before HIFU, the contents of γδT cells in blood of rats in group 1 and 2 were detected by flow cytometry(FCM), and at day 1,3,5,7 after treatments, the contents of γδT cells in group 3, 4, 5 were detected by the same assay. Meanwhile, HSP70,60,27 in tumor tissues of rats in group 2, 3, 4 were detected by Western blot. Results: 1. UMR106 cells can induce sarcoma with numerous osteoid and intending to occur pulmonary metastasis spontaneously. 2. γδT cells increased in 7 days after HIFU, and reached the peak at day 3 with 249.08% amplification. And at day 7, γδT cells recurred to the untreated level. 3. After HIFU, HSP70 in tumor tissues increased , and arrived the peak at day 3, and had kept higher than untreated group until day 7 and HSP27 increased at day 5. While no change of HSP60 was detected. 4. While the increases of HSP70 and 27 were inhibited by QCT, the content of γδT cells were also decreased in group 4, while no changes of γδT cells were observed in rats with QCT only. Conclusion: γδT cells in blood of rats with osteosarcoma increased significantly at the early stage after HIFU treatment . And this change is related at least partly to the increased expression of HSP70 and HSP27.
