OBJECTIVES: To study the effect and antitumor mechanism of Resveratrol combine 5-FU on Human gastric cancer cell lines SGC7901 and BGC823 in vitro and its mechanism of action. METHODS: MTT assay was used to analyze the effect of Resveratrol and 5-FU alone or in combination on the growth of SGC7901 and BGC823 cells. Transmission electron microscopy , Flow cytometry and immunocytochemical staining were used to observe the morphologic changes, cell cycle and expression of Survivin,Caspase-3,Fhit,Pcna of these cells. RESULTS: 1.Alone or in combination Resveratrol and 5-FU could inhibit the growth of SGC7901 and BGC823 cells in a time and dose dependent fashion. The combination of resveratrol with 5-FU had synergistic effect on the SGC7901 cell line when the inhibition rate exceed 85% and on the BGC823 cell line when the inhibition rate exceed 65%. 2.SGC7901 and BGC823 cells presented some morphologic features of apoptosis after treatment with Resveratrol and 5-FU for 72 hours. 3.Resveratrol could arrest cell cycle of SGC7901 and BGC823 cells in G0/G1 phase. 4.Resveratrol and 5-FU could downregulate the expression levels of Survivin,Pcna,upregulate the expression levels of Caspase-3,but the levels of Fhit is not change. CONCLUSION: 1.Alone or in combination Resveratrol and 5-FU could inhibit the growth of SGC7901 and BGC823 cells effectively .The combination of them showed synergistic or antagonistic effects at different concentration. 2.Resveratrol can hardly change the distribution of cell cycle. 3.Resveratrol can make gastric carcinoma presented morphologic features of apoptosis. 4.Resveratrol may induce apoptosis in SGC7901 and BGC823 cells by reducing the expression levels of SVV,Pcna, upgrade the expression levels of Caspase-3,have nothing to do with levels of FHIT.
