Objective To investigate the relationship between a common functional polymorphism in promoter of microsomal triglyceride transfer protein gene (MTP-493G/T) and the levels of serum lipids as well as apolipoproteins in Guangxi Hei Yi Zhuang population. Methods A total of 500 people of Hei Yi Zhuang nationality were surveyed by a cluster sample. Blood pressure, body height, body weight, serum lipid and apolipoprotein levels were measured, and both body surface areas and body mass index were calculated. The genotype and allele of MTP-493G/T were determined by the polymerase chain reaction-allele specific oligonucleotides (PCR-ASO), then confirmed by direct sequence measuring. The results were compared with those in 500 people of Han nationality who also live in that district. Results (1)Allele frequency The frequencies of G and T allele were 0.74, 0.26 in Hei Yi Zhuang people; and 0.73, 0.27 in Han people; respectively. The frequencies of MTP-493 G/T genotype were GG, 0.58(289/500); GT, 0.33(166/500); and TT, 0.09(45/500) in Hei Yi Zhuang; and GG, 0.56(281/500); GT, 0.34(172/500); and TT, 0.09 (47/500) in Han people; respectively. The frequencies of G/T allele and genotype of MTP-493 were not significantly different between the Hei Yi Zhuang and Han people. (2)Effects of MTP genotypes on lipids and apolipoproteins The levels of TC, TG, LDL-C, and Apo B were very similar between the GG and GT genotypes (TC: 4.51±1.56 vs. 4.57±1.75 mmol/L; TG: 1.55±0.65 vs. 1.56±0.55 mmol/L, LDL-C: 2.23±0.63 vs. 2.28±0.72 mmol/L; Apo B: 0.78±0.47 vs. 0.81±0.21 g/L; P>0.05 for all). However, the levels of TC, TG, LDL-C, and Apo B in TT genotype were significantly higher than those in GT or GG genotype (TC: 4.97±1.83 mmol/L,P<0.05 with GT or GG genotype; TG: 2.31±1.31 mmol/L,P<0.01 with GT or GG genotype; LDL-C: 2.51±1.23 mmol/L,P<0.01 with GT or GG genotype; Apo B: 0.95±0.37 g/L, P<0.01 with GT or GG genotype). The levels of HDL and ApoA1 were not significantly different among the three genotypes. Conclusions The present study showed that the frequencies of allele and genotype of MTP-493G/T were not significantly different between Hei Yi Zhuang and Han population. But the levels of TC, LDL-C, TG, and Apo B in TT genotype were significantly higher than those in GT or GG genotype in both nationalities. The levels of HDL-C and ApoA1 were not significantly different among the three genotypes.
