Objective To investigate the expression of tumor necrosis factor alpha(TNF- α ) in subcutaneous and omental adipose tissues of overweight and the relation with body mass index(BMI), waist-hip ratio(WHR ), lipids, fasting serum insulin(FINS ), fasting serum glucose(FBG)and insulin resistance index(HOMA-lR). Methods Total mRNA was extracted from adipose tissues of AnHui Chinese. Compared with the expressions of β -actin gene , The TNF- α expression level was measured by semi-quantitative RT-PCR in abdominal subcutaneous and omental adipose tissues of 24 normal controls (18.5kg/m2 ≤BMK24kg/m~2) and 19 overweight(24kg/m2 ≤ BMI<28kg/m2 ).Anthropometric data such as height ,body weight and waist to calculate body mass index and waist-hip ratio. Systolic blood pressure(SBP) and Diastotic blood pressure(DBP) were also measured. Fasting serum insulin, glucose, lipids were determined. Insulin resistance index were caculated .Than using t-test, ANOVA, linear correlation analysis, multiple factors stepwise regression for statistical analysis Results The expression of TNF- a mRNA in subcutaneous and omental adipose tissues were not significantly different in overweight and in controls(p>0.05). There were no differences in the expression of TNF- α mRNA between subcutaneous and omental adipose tissues from each subject(p>0.05). WHR, WC, SBP, DBP, TG, VLDL-C , FINS, HOMA-IR were significantly higher in overweight than in normal (p<0.05). The expression of tumor necrosis factor alpha in subcutaneous and omental adipose tissues was found to have significant positive correlation with WHR, WC, FINS, HOMA-IR (p<0.05).
