Background: The pathogenesis of congestive heart failure has been studied as an important problem for many years. The ventricular remodeling includes not only the myocardial cell but also the cardiac interstitial collagen. Nuclear factor kappa B(NF-kB) is an inflammatory transcription factor ,the relationship between it and congestive heart failure is becoming research focus .Tumor necrosis factor-a(TNF-α) is a proinflammatory cytokine that been implicated in the pathogenesis of cardiovascular diseases, including acute myocardial infarction, chronic heart failure , atherosclerosis ,viral myocarditis and cardiac rejection. Angiotensin converting enzyme inhibitor (ACEI) can improve cardiac function and inhibit ventricular remodeling and myocardial fibrosis. The prostaglandin has the effect on inhibition the cardiac interstitial collagen, which is one of the functions of ACEI. Many studies have recently acknowledged that there is a close relation between heart failure and TNF-α, NF-κB,BNP. This study would provide new insights and important therapeutic bases preventing congestive heart failure.Methods: Seventy adult male Wistar rats weighing 250-300 g were used in the study. Congective heart failure was produced by ligation of the left anterior descending coronary artery in the rats.24 hours after myocardial infarction ,rats were randomized to untreated infracted group(CHF) or ACEI group (Cilazapril 1mg·kg-1·d-1).Sham-operated rats served as control group .After two weeks ,these rats were sacrificed. The weight of rats body and heart were measured .The activities of plasma Ang II were examined by the method of radioimmunoassay lest .The gene expression of cardiac TNF-α and BNP were measured by reverse transcription-polymerase chain reaction (RT-PCR) method. We used ELISA to detecet the circulatin level of myocardial TNF-α ,and used EMSA to detect NF-κB.Results: ①There was no significant difference in the body weight among the three groups. ②Heart weight, as well as the ratio of heart weight to body weight in the CHF group were significantly increased compared with control...
