Objective To study the effect induced by arsenic trioxide(As2O3) alone and combinated with cisplatin( DDP )on human cervical carcinoma Hela cells .Methods Hela cell line was treated by As2O3 or DDP or both.Cell proliferationwas evaluated by MTT assay and cultured colony formation method. Cell morphologic changes were examined using light microscrope , Giemesa-wright' s dye and acridine orange dye. Flow cytometry was used to examine apoptosis on cervical carcinoma Hela cells . The expression of Bcl-2 protein and Bax protein were investigated by immunohistochemical techniques.Result MTT assay showed that arsenic trioxide inhibited human cervicalcarcinoma Hela cells growth in vitro in a dose and time-dependent manner (P< 0.05 ). Cultured colony formation method showed that colony formation treated with AS2O3 reduced compared with that of controls (P<0.05) . Apoptotic cell morphologic changes were examined using light microscrope , Giemesa-wright' s dye and acridine orange dye. Flow cytometry showed that AS2O3 induced the apoptosis in a dose and time-dependent manner (P < 0.05) . When treatment with combination of AS2O3 and DDP, the inhibitory effect on proliferation , colony formation and apoptotic rate were markedly enhanced (P < 0.05) .conclusion 1, Arsenic trioxide can significantly inhibit Hela cells growth in...
