OBJECTIVE: This study was conducted to investigate the injuring effect and mechanism on human umbilical vein endothelial cells (HUVECs) induced by formaldehyde, in order to elucidate the role of endogenous formaldehyde in the development of cardiovascular disease which its endothelium function was obstructed such as atherosclerosis and vascular complication of diabetes.METHODS: HUVECs-12 were exposed to various concentrations offormaldehyde (0, 5,10, 20,40, 80μmol/L) for 24 hours as experimental group, treated by hydrogen peroxide and anti-oxidant medicine vitamin C as positive control group and protective group respectively at the same time; In order to observe time-effect of formaldehyde, HUVECs-12 were treated with 40μmol/L formaldehyde for 0, 4, 12, 24, 48, 72 hours respectively. Morphological change of cells was observed by H-E staining; The viability of HUVECs-12 was determined by a colorimetric 3-[4, 5-dimethyl thiazol-2-yl]-2, 5-diphenyltetrazolium bromide (MTT) assay; The transudation content of LDH was observed by colorimetry reaction; The level of MDA in supernatant-conditioned media of cells was measured by thiobituric acid reaction (TBA) method.; The nitrite content of the culture medium was measured by nitrate reductase method; The activity of NOS was measured by biochemical approach; The protein and mRNA expression of eNOS and iNOS of HUVECs was detected by immunohistochemistry and RT-PCR approach respectively.RESULTS: Formaldehyde induced cell morphological change, inhibited theviability of cells, increased the transudation content of LDH on HUVECs-12 in concentration-and time-dependent manner; Formaldehyde increased the level of MDA and NO in supernatant-conditioned media of human umbilical vein endothelial cells in concentration-dependent manner, reaching the top at 24 hours after stimulation;...
