OBJECTIVE To explore the influence of recombinant human growth hormone (rhGH) on apoptosis of T lymphocytes subgroubs and IgA plasmocytes of intestinal wall of the gut immune barrier during the gut barrier injury.METHOD 60 Wistar rats were divided into two groups randomly. One is experiment group which used rhGH, the other is control group, in addition , each group was divided into three subgroups (2,4, 6days, respectively). Each subgroup has 10 Wistar rats. All rats were made into ischemia-reperfusion model. The rats of experiment group were injected rhGH (1.33u/kg/d) on postmodel everyday. The changes of apoptosis of CD8+, CD4+, CD3+ T lymphocytes and IgA plasmocytes of ileal mucosal and laminapropria of the distal end of ileal mucosal tissue were observed by immuohistochemical method .RESUAL (1) On the 6th day the apoptosis of CD3+ T cell and IgA plasmocytes in the control group is markedly increased compared with on the 2nd day and the 4th day(P=0.001, P=0.000),but on the 2nd day and the 4th day there are no difference. The apoptosis of CD8+, CD4+T cell have no difference on the 2nd and 4th and6th day. On the 6nd day the apoptosis of CD8+, CD4+ T cell in experiment group are less than they are on the 4th day and have obvious difference(P=0.001, P=0.000). On the 6nd day the apoptosis of CD3+ T cell and IgA plasmocytes in experiment group are more than they are on the 4th day and the 2nd day and have obvious difference (P=0.000, P=0.000) . There are no difference on the 2nd day and the 4th day . On the 2nd and 4th and6th day the apoptosis of IgA plasmocytes are gradually decreased and have obviously difference between the control and the experiment group (P=0.005, P=0.011, P=0.000) . On the 2nd day the apoptosis of CD8+, CD4+, CD3+ T lymphocytes are gradually decreased and have obviously difference between the control and theexperiment group (P=0.019, P=0.013, P=0.002).(2) Morphologic observation; On the 2nd day experiment group showed intestinal villus hyperplasia and intestinal gland hyperplasia compared with control group.On 4th day experiment group showed the serious inflammation of intestine mucosa. Meanwhile intestinal villus showed hyperplasia obviously. On 6th day experiment group showed intestinal mucosal epithelial cell and villus hyperplasia obviously and intestinal gland hyperplasia regularly.CONCLUSION During the period of gut barrier injuried, apoptosis of T lymphocytes subgroubs and IgA plasmocytes of intestinal mucosal and laminapropria of the gut immune barrier are increased.using rhGH not only improves the mechanical barrier of intestinal mucosal in morphological structure but also obviously inhabit the apoptosis of intestinal Immunologic cell, the impact on IgA plasmocytes is obvious in particular. The probable time of action maybe is on the 6th day markedly.
