Objective: To investigate the antitumor efficacy of combining endostatin with doxoiubicin and explore its intrinsic mechanism ,the effects of endostatin on healing of incision and liver regeneration in mice.Methods: First section. Hepatocellular carcinoma graft was implanted in forty mice after resecting left liver lobe ,and were randomized into 4 groups to received every other day for two weeks before sacrifice peritoneal cavity injection of: 1 .control group(nomal saline), 2.Es group(12mg/kg), 3.Adm group(lmg/kg), 4.combined group(Es 12mg/kg , Adm lmg/kg).Tumor volumn ,intratumoral microvessel density(MVD),apoptotic index(AI) and liver function were evaluated.Second section. Total mice were sacrificed after two weeks.the influence of endostatin on healing of wounds and liver regeneration was studied by comparing MVD,light microscope pictures and electon microscope pictures in different groups.Results : 1.First section.The antitumoral effects of combined group were siginificantly different from Es group or Adm group(p<0.05).There was difference between Es group and Adm group.The liver function of Es group is similar to control group(p>0.05),Whereas the liver function of Adm group and combined group was remarkable different from control group(p<0.05).2.Second section.There was no difference in the MVD.contribution and density of fiber and liver regeneration in Es group versus the other groups.Conclusions: 1.There were synergistic antitumoral effects by combining Es withAdm.The inhibitory effects of Es were not only better than of Adm but also no toxicity to liver,specially suitable to be used early under liver disfunction.2.Es has no adverse effects on the healing of incision and liver function.The methanism of targeting tumor is to be elucidated.
