| Objective: To obtain satisfied startup information related to carotid artery restenosis in canine model by ballon angioplasty. Material and methods: Ballon angioplasty was performed on 12 dogs. PDGF,IGF,FGF,MAC-1,LFA-1 in venous blood were measured on the point of pre-operation and 2, 4, 6, 12, 24, 48, 72 hours after operation, and DSA was performed on 2 dogs every 2 weeks on the point of 2, 4, 6, 8, 10 weeks after operation in order to measure the rate of stenosis. At the same time, the dogs were sacrificed and the target vessels were excised. For morphologic examinations the tissue sections were stained with HE, VSMCs were indicated immunohistologically with an antibody against α-acting, VSMCs proliferation was examined by PCNA, and apoptosis was examined by TUNEL. Through statistic analysis and pathology verification, some satisfied results were obtained. Results: Carotid artery restenosis models were established in 10 dogs. IGF release started up at 4 ~6 hours after operation (average 942.55±44.13ng/ml, 1073.09±87.73ng/ml), the peak value arrived at 12 hours after operation (average 1468.35±101.85ng/ml) and maintaind till 72 hours (average 1405.64±85.09ng/ml); MAC-1 release started up at 12 hours after operation (average 13.09±1.31%), the peak value arrived at 48 hours after operation (average 22.39±3.92%), and maintained till 72 hours (average 18.78±3.81%); FGF only released briefly at 12 hours after operation (average 12.35±1.45pg/ml) at low value; PDGF and LFA-1 did not release in 72 hours; the rates of stenosis peaked at 8 weeks after operation (average 62.76±3.03%); the tissue sections stained with HE displayed vessels stenosis , neointimal formation, which contained predominantly large shuttle-shaped VSMCs as detected by α-acting staining; the counts of PCNA positive cells peaked at 4 weeks after operation (average 61.60±1.96); the amount of apoptosis cells decreased in the neointimal of 2,4 weeks, situation of 6,8,10 weeks after operation and normal was on the contrary. Conclusion: Carotid artery restenosis from CAS can reappear in canine model by ballon angioplasty, IGF and MAC-1 are wanted startup information related to carotid artery restenosis, VSMCs proliferation and migration occur mostly in 4 weeks after operation, and stenosis form stably at 8 weeks. |
