Recent studies have demonstrated that the coronary heart disease (CHD) has become a very common and familiar disease which has serious threaten old people's health and lives .Passing by more than 20 years of the clinical development practice and experience accumulate, the percutaneous stransluminal coronary angioplasty ( PTCA) having become the extensive treatment choice for more and more coronary sufferers because of its convenient operation, high success rate and a little complications.However, the restenosis (RS) rate is up to 32%-57% in 3-6 months after the PTCA in the rat carotid artery, this obstructe its application and development on the clinic seriously.Modern study tested that the RS after PTCA is a many links and continuous organization respond process.For the last 20 years, many of people have been carrying on all-directions, multilayers of foundation and clinic research for preventing and treating RS after PTCA, but the total result has been not fulfil person's expectation.As to the pathobiologic process of blood vessel restenosis following balloon angioplasty, traditional Chinese medicine (TCM) believe that the RS belongs to the category of " stagnation of blood symptom", the basic pathogenesis of RS is Ben deficiency and Biao excess.The main featue of Ben deficiency is deficiency of zang-Qi,besides deficiency of Qin and deficiency of Yang;The major feature of Biao excess is stagnation of blood, besides internal retention of phlegm and water, stagnation of Qi and cold stagnation. So supplementing Qi and promoting blood circulation are the main therapeutic principles for preventing RS after PTCA. TCM have affluent Bianzheng Lunzhi system and concept of holish and Chinese traditional medicine compound have various chemical components and extensive biology activitives, which provide the possibility on theory for holding up the pathology and physiology characteristics of RS after PTCA .ObjectionBased the theory of Qi and blood in TCM and combining the key pathogenesis link of RS, Three Chinese herbal medicines:sanqi, huangqi, chuanxiong ,which have major function of reinforce Qi, promoting flow of Qi and promoting blood circulation to remove blood stasis, were chosed to compose compound prescription.Then we adopt quantitation of tissue method combining the pathologic morphology analyse to comprehend the further occurrence mechanism of RS.The study investigate the protective effect and explore the protective mechanism of yiqihuoxue prescription,which can provide scientific proof to enlarge the clinical application of the same kind of Chinese herbal medicine and the new preparation of RS after PTCA.Thus may develop a new path to solve RS.Methods48 male Sprague-Dawley big rats were randomly divided into 4 groups (n=12):the sham-operated group (A),the model group(B), the low-dose yiqihuoxue prescription group (C), the high-dose yiqihuoxue prescription group(D).Each group included 2 time points(5 days and 28 days)(n=6) .The balloon catheter injury was perfomed on left common carotid artery of big rat by imitating the document[3].Each group of Chinese herbal medicine were given to 6 rats (0.8ml/100g/d) from lday before balloon injury to 5day or 28day afterballoon injury.Namely,the sham-operated group and the model group were similarly treated with 0.8ml/100g/d of saline.Around 6 rats from each group were euthanized( 3% Pentobarbitalum Natricum 30mg/kg) by exsanguination at appropriate time point.The injuried left common carotid arteries were carefully removed and harvested for specific protocols. The HE staining for imaging analysis ,the TR-A02 elasticity fiber staining for elastin and Masson's staining for collagen and VSMC were carried out to assess the effect of yiqihuoxue prescriptions .Microscopic analyses and quantitation of morphologic parameters were performed using HMIAS-2000, which is a full-automatic computerized colorful imaging analysis system.
