The expression of three important proteins of Rho GTPases was studied in hepatoma cell lines and the significance of GTPase in the occurrence and development of hepatoma was also investigated. Protein expression levels of Racl, Cdc42 and RhoA in several hepatoma cell lines were examined by Western-blot. Compared with normal liver cell line, protein expression levels of Racl, Cdc42 and RhoA in three hepatoma cell lines were up-regulated, which implied that Racl, Cdc42 and RhoA might play an important role in the development and progression of hepatoma.In further research, the small interference RNA technique was employed to knock down gene expression of Racl in hepatoma cell line 97-H. The protein level of Racl, the cell proliferation, the cell vitality and the ability to invade the reconstituted basement membrane were decreased dramatically after transfection with a Racl-specific siRNA vector .The results suggested that Racl might play an important role in the development, progression, transference of hepatoma and also provide a possible strategy for the interference of tumor angiogenesis by targeting the Racl.
