In this dissertation, the extraction and purification of polysaccharides from Epimedium were systematic studied. A high molecular weight polysaccharide mono-fraction was purified, and some physicochemical characters and structural properties of this fraction were researched. The work involved are as follows:1. The method of content determation including Epimedium polysaccharide, protein and uronic acid was established. Based on these methods, the orthogonal test of four factors and three levels was studied to obtain the extracting technology of epimedium polysaccharide. The finally rational technology condition were: 100 grams epimedium powder was soaked with 1500mL water, then extracted at temperature of 100 ℃ and three times for 1.5 hours. Following this condition, the yield of polysaccharide was 7.78%.2. Macroporous adsorption resin was adopted to decolorize and deproteinize from crude Epimedium polysaccharide, Influence factors, such as pH value, Tannic Acid, elution volume et al, were investigated. The content of proteins in crude Epimedium polysaccharide decreased from 1.2% to 0.035%, and the colour of the product is grey. The results showed that deproteinization and decolo -urization from crude Epimedium polysaccharide by macroporous adsorption resin is a feasible method,3. After desalting Epimedium polysaccharide, DEAE A-25, DEAE Sepharose CL-6B, Sephacryl S-200HR, Sephacryl S-400HR were used respectively to separate and purify Epimedium polysaccharide. HPCE and HPGPC were used to determine the purity and the molecule weight of ten parts obtained. GC and NMR spectrometry investigation of EPSBI2 which was one of ten parts revealed a →4) a -D-GalpA(1 → 4)- a -D-GalpA(1→ repeating unit.4. The immunocompetence of ten parts were determined, the results showed: EPSAI1, EPSAI22, EPSAⅡ和EPSBⅢ3 have excellent immunocompetence.
