Therefore more and more people pay attention to CA.There were various kinds of study of CA before, mainly the detection of HPV. The abnormal proliferation of Keratinocytes caused by the infection of HPV, the change of cellular immune function and the exploration of treatment methods for CA. CA is a kind of benign papilloma proliferation disease, And recently some research also indicate that the pathogeny of over proliferation of Keratinocytes caused by the infection of HPV is the increase of cell split and the reduce of death of cells infected by HPV. Survivin has recently been identified as a novel inhibitor of apoptosis (IAP). It is expressed in many common human tumors, Survivin mainly blocks cell apoptosis by directly inhibiting terminal effector caspase-3 and caspase-7 activity, and it has also positive correlation with cell proliferation, blood vessel formation; Bax is the protein promoting apoptosis widely researched in Bcl-2 family, its expression descends in many tumors, the expression is increased in many cell apoptosis; Ki-67 is a proliferating cell nuclear antigen, playing a great role in regulation of cell cycle, its rate of expression canreflect the state of cell proliferation rightly. At present, the expression of Survivin mRNA and Bax protein in CA and their correlations have not been reported at home and abroad. This study was designed to explore its expression in CA and their correlations to investigate the relationships between the pathogeny of CA and survivin ,Bax, Ki-67, and explore the mechanism of CA.Materials and methods1.MaterialsThe vulva tissues of 38 cases were obtained from the patient of CA of theout-patient department of Dermatology and Venereology of the First Affiliated Hospital of Zhengzhou University, all the tissues were required to accord with the principles: particular case history and feedback record; therapy out of medicine , laser, or immunity; the pathologic confirmation.the test of acetic acid soaking is positive, infected by HPV6/11 approved by detected with in situ hybridization. The penis tissues of normal adult man were obtained from the patient from urology surgery in the first affiliated hospital of zhengzhou university,the pathologic confirmation. The tissues is normal skin were confirmed by pathology. 2.MethodsThe situ hybridization and immunohistochemical methods were respectively used to detect the expression of Survivin mRNA,Bax and Ki-67 protein in CA and normal skin cases. The relationship between the expression of Survivin mRNA and Bax or Ki-67 protein in CA cases were also systematically analyzed. 3.Statistical analysisThe SPSS statistical package program 11.0 was used for all analyses. Associations between the variables were tested by x~2 test and Fisher's exact test of probabilities, t test, and Pearson correlation. Satistically significant level was considered as "alpha equals 0.05".Results1. The expression of Survivin in CA tissuesThe positive expression of Survivin mRNA were mainly localized in the cytoplasm.the positive cell were mostly concentrated in the upside and middleside of the prickle cell layer and stratum granulosum. The colour of the positive signal mostly stain light yellow,palm fibre and snuff color, minority stain the downside of the prickle cell layer and basal cell layer. The positive rate of Survivin mRNA were 68.4%(26/38), 0.00%(0/15)in CA and normal skin tissue respectively, the expression of intensity is 3.921±0.254, 0.883±0.088 respectively. Compare with normal skin tissues,, there were significant difference in the expression of Survivin mRNA between in the CA and normal skin tissues(P<0.01).2. The expression of Bax in CA tissuesThe positive expression of Bax protein were mainly localized in the cytoplasm. The positive cell were mostly concentrated in the upside and middleside of the prickle cell layer and stratum granulosum. there are few expression in the downside of the prickle cell layer and basal cell layer. The colour of the positive signal mostly stain light yellow,palm fibre and snuff color, minority stain palm brown. The positive rate of Bax protein were 76.3%(29/38),66.7%(10/15)in CA and normal skin tissue respectively, the expression of intensity is 3.813±0.246,2.961±0.244 respectively. Compare with normal skin tissues, there were no significant difference in the positive rate of Bax protein between in the CA and normal skin tissues(/)>0.05),but there were significant difference in the expression of intensity of Bax protein between in the CA and normal skin tissues(P<0.01).3. The expression of Ki-67 in CA tissuesThe positive expression of Ki-67 protein were mainly localized in the nucleus. The positive cell were mostly concentrated in the downside of the prickle cell layer and basal cell layer, there are few expression in the upside and middleside of the prickle cell layer and stratum granulosum. The colour of the positive signal mostly stain palm fibre and snuff color, minority stain palm brown. The positive rate of Ki-67 protein were 84.2%(32/38), 60.0%(9/15)in CA and normal skin tissue respectively, the expression of intensity is 0.234±0.021, 0.05±0.006 respectively. Compare with normal skin tissues, there were no significant difference in the positive rate of Ki-67 protein between in the CA and normal skin tissues(P>0.05).but there were significant difference in the expression of intensity of Ki-67 protein between in the CA and normal skin tissues(f><0.01).4. The correlation analyse of the expression of Survivin, Bax and Ki-67.The results of analysis of correlation of the expression in CA indicate that Survivin has significant positive correlation with expression of Ki-67(r= 0.497, P<0.01), but has significant negative correlation with expression of Bax(r =-0.505, /><0.01).Conclusionsl.The over expression of Survivin mRNA in the CA tisseus and The expression of Survivin in CA tisseus has significant positive correlation with expression of Ki-67 suggest that the significance of survivin mRNA expression is related to restrain the keratinocytes apoptosis or promote its abnormal proliferation.2.The over expression of Bax in the CA tisseus suggest that it is the response to inhibit theinfection of HPV.3.The expression of Ki-67 in CA tissues is higher than its expression in normal skintisseus indicate that the formation of CA is related to over cell proliferation caused by theinfection of HPV.4.The expression of Survivin in CA tisseus has significant negative correlation withexpression of Bax suggest that Survivin may resistant the expression of Bax or Baxpromoting apoptosis function, furthermore through whick take part in the formativemechanism of CA. | | Keywords/Search Tags: | condyloma acuminatum, Survivin, Bax, Ki-67 | | Related items |
