AB:Objective: (1) To explore a better method of establishing rat model ofcombined liver and kidney transplantation. (2) to investigate the relationship betweenthe Ragulated upon Activation Normol T Cell Expressed and Secreted(RANTES)andacute rejection of combined liver and kidney transplantation in rats.Methods:SD rats and Wistar rats were randomly divided into normal group, isograftgroups, and allograft group. The model of combined liver and kidney tranplantation inrats was set up by blood vessels reconstructed with cuff methods. Liver and kidneybiopsies from the recipients were abtained on the day 1, 4, and 7 postoperatively toexamined the expression lever of RANTES in liver and kidney grafts. Usingimmunohistochemmistry stain, samples were firstly stained and then applied to imagesemiquantitive analysis.Results: (1)The model of combined liver and kidney transplangtation in rats was setup successfully, The allograft group all died 9 to 14 days after transplantation , andtheir histopathologic examination showed typical allografts rejection., while theisograft grafted survived more than 90d. (2)The expression lever of RANTES inallograft group was obviously higher than those in both isograft group and normalgroup. Moreover, the expression lever of RANTES was correlated positived with thedegree of acute grafts rejection.Conclusions: (1) This rat model of simultaneous liver-kidney transplantation issimple and reliable, and donor liver and kidney of Wistar rats and recipient SD ratscombination is a high rejection model which can be effectively used to study allograftrejection. (2) Detecting the expression lever of RANTES in liver and kidney graftstissues may be helpful to the diagnosis of acute rejection and the judgement of thedegree of acute rejection.
