Objectives The result of modern molecular biology investigationdisplay,meny cytokine could accelerate growth new angiogenesis, they facilitate wound plerosis , increasing skin flap survival rate.Before long nerve growth factor ( NGF) was investigated in nervous system regeneration and plerosis,but NGF was pay close attention to soft tissues recently.Nerve growth factor could facilitate fibroblast and vessel endothelial cell proliferation. acceleration inflammatory reaction. some research reports play a part in wound healing.Whether the NGF effect skin flap survival ? Thereby,the purpose of the present study was to determine the effects of nerve growth factor on survival of the full skin flap in rats. In addition,discussed some probable way that may contribute to this effect.Methods A dorsal random skin flap model (2×8cm~2) was used in thisstudy. Twenty male Sprague-Dawley rats were randomly divided into two groups(N1=10, N2=10).Reconstitute murine natural 7s NGF (0.5ug) wasinjected into subdermally the flap at 2, 4 and 6 cm distances from the distal pedicel 10 rats, at preoperative 24 h, operative and postoperative 24h three times. Flap that received a saline injectionwere used as the controls.The skin flap survival an area of was measured on postoperativeday seven.Results1 The flap mean viable surface area for the control group was 5.571±0.691 cm (7th day). The group with injection of NGF had a mean viability of 7.581±1.48 cm 2 (7th day) . Statistically significant differences were obtained in treatment groups in comparison to the control group. (p=0.002<0.01).2 There was significant difference in blood stream worth dorsal random skin flap of rats treated with NGF compared to the control group.3 The microvascular density(MVD) in the experimental group (14.3 ±2.8 ) was higher than in the control group (11.12+2.9) . (P=0.017 < 0.05) .Conclusion This is the first time to elucidate that administration of exogenous NGF could increase flap blood stream significantly enhance survival an area of flaps, It may possibly throughinduction of vessel growth to get this effect.
