objective:To investigate the changes of activities of hexokinase (HK), malate dehydrogenase (MDH),lactate dehydrogenase (LDH) and glucose-6-phosphate dehydrogenase (G6PD) in oesophageal squamous cell carcinoma compared to oesophageal normal tissue. The approach to take energy in oesophageal squamous cell carcinoma was also anlysised. In addition, we studied the relationship between glucose metabolic enzymes activities and diifferent T stages, lymph node metastasis and weight loss.Methods:Activities of HK,MDH,LDH and G6PD were detected by colorimetry in 30 oesophageal squamous cell carcinomas and oesophageal normal tissues. Activities of the enzyme were also studied in oesophageal squamous cell carcinoma in diifferent T stage, with or without lymph node metastasis and weight loss.Results:(l)The enzyme activities of HK, MDH, LDH and G6PD were 6.495 U/gprot, 139.95 U/gprot , 6227.6 U/gprot and 16.2 U/gprot respectively in oesophageal squamous cell carcinomas;and were 4.124 U/gprot, 112.28 U/gprot, 4683.3 U/gprot and 8.7 U/gprot respectively in normal oesophageal squamous cell. The activities of HK , MDH, LDH, G6PD in oesophageal squamous cell carcinomas were significantly increased compared to those in normal tissues(p<0. 01).(2) The activities of HK in Tl, T3andT4 of oesophageal squamous cell carcinomas were 5.542 U/gprot, 6.124 U/gprot and 6.959 U/gprot respectively; MDH were 133.82 U/gprot, 141.05 U/gprot, 145.84 U/gprot respectively; LDH were 5707.0 U/gprot, 6141.9 U/gprot> 6790. 2 U/gprot respectively; and G6PD were 8. 85 U/gprot, 15.5 U/gprot, 24.2 U/gprot respectively. The activities of the enzymes in oesophageal squamous cell carcinomas were significantly increased compared to those in normal oesophageal squamous cell, while the activities of HK and MDH were not correlated with tumor stage (T1-T4) (P<0.05). The activities of G6PD and LDH were correlated with different T stages (T1-T4)(P<0. 05) (3) The activities of HK, MDH, LDH and G6PD were 6. 124 U/gprot, 123.5 U/gprot, 6242.0 U/gprot, 16.1 U/gprot respectively, if the lymph node were normal. They were 7.939 U/gprot, 149.80 U/gprot, 7352.7 U/gprot, 28.7 U/gprot respectively, if lymph node were metastasized by cancer cells. Activities of HK , MDH , LDH andG6PD were not correlated with lymph node metastasis in oesophageal squamous cell carcinomas(P>0.05).(4) The activities of HK> MDH> LDH and G6PD were 6.241U/gprot, 133. 63 U/gproU 6227. 6 U/gprot> 15. 27 U/gprot respectively, if the weight was not lost . They were 7.352U/gprot^ 154.70 U/gprot> 7634. 5 U/gprot> 19. 65 U/gprot respectively, if the weight was lost. Activities of HFU MDH ^ LDH> G6PD were not correlated with weight loss(P>0. 05).Conclusion:(l)The activities of HK, LDH, G6PD were increased in oesophageal squamous cell carcinoma, which indicate the greater ability to take glucose in oesophageal squamous cell carcinoma. Aerobic and anaerobic metastasis were also increased in oesophageal squamous cell carcinoma.(2) Along with the progress of T stage , the activities of LDH increased, which suggest the progress of oesophageal squamous cell carcinoma was associated with higher anaerobic metabolism.(3)Activities of aerobic and anaerobic enzymes were not correlated with lymph node metastasis or weight loss.
