[Objective] To explore the relationship between gallstone formation and the serum of apolipoprotein E concentration as well as gene polymorphism. [Methods] Seventy-eight patients with gallstone and forty-three non-gallstone in Department of general surgery of West China Hospital from Oct 2004 to Mar 2005 were gathered. The study identified apoE genotypes by the technique of polymerase chain reaction-restriction fragment length polymorphism.The serum of apolipoprotein E concentration of different genotypes were compared and analyzed the relationship with gene polymorphism and gallstone formation.[Result] 1. The prevalence of genotype E3/ 4 was 23 %(9/ 40) in the cholesterol gallstone group , which was significantly higher than that of 7 %(4/ 60) in the control group (P < 0.05). The prevalence of allele ε 4 was 15 % (12/ 80) , which was significantly higher than that of 8 % (9/ 120) in the control group ( P < 0.05) .2.The total cholesterol and high density lipoprotein-cholesterol of gallstone group was marked differently with non-gallstone group and the serum concentration of TG , TC and HDL of genotype E3/ 4 of Apo E was marked differently with other genotypes.3.Thelevel of apolipoprotein E serum concentration of gallstone group was increased significantly compared with non-gallstone group. The level of apolipoprotein E3/4 serum concentration increased significantly compared with other genotypes(P<0.05).4. The level of apolipoprotein E serum concentration had positive correlation with TG and TC and had negative correlation with HDL.Conclusions 1. Genotype E3/ 4 of ApoE may contribute to the development of gallstone in Chinese and allele e 4 seemed to be a genetic susceptible factor in gallstone formation.2. The results suggest that the average level of serum lipids in the same apoE phenotype patients with gallstones is higher than that in controls , and the different apoE phenotypes patients with gallstones have different characteristics of dyslipidemia.3.The higher serum of apolipoprotein E concentration may contribute to the risk factors development of gallstone.4.The level of serum of apolipoprotein E concentration had positive correlation with TG,LDL and TC and had negative correlation with HDL.
