Expression Of CD147,MT1-MMP Protein And Its MRNA In Gastric Carcinoma And Their Significance

Objective: To investigate the expression of the protein of CD 147 and MTl-MMP , and mRNA of the latter in normal gastric and carcinoma tissues and find out their correlations and their relationship with the biologic behavior of gastric carcinoma. Methods: CD147 and MTl-MMP protein and its mRNA were detected by immunohistochemical staining and in situ hybridization (ISH) respectively in 10 cases of normal gastric tissue and 59 cases of gastric carcinoma tissues. Results: The expression level of both CD 147 protein and MTl-MMP protein in normal gastric tissues was significantly lower than in gastric carcinoma tissues(P<0.05).These protein was mainly stained on the cytoplasm and (or) cellular membrane. The expression of MTl-MMP and CD 147 protein in the subgroup of bad-differentiated with the invasiveness depth of tunica muscularis and tunica serosa and lymph nodes metastasis was significantly higher than the other subgroup of well-differentiated with invasiveness depth of tunica mucosa and tela submucosa and without lymph nodes metastasis (P<0.01). The expression level of MT1-MMPmRNA in gastric normal tissues was also significantly lower than in carcinoma tissues(P<0.05).If judgd with the expression of the cancer cells, there was not significant difference between the condition of differentiation , the depth of the invasiveness and lymph node metastasis (P>0.05). MTl-MMPmRNA could be seen on carcinoma cells and stromal cells and it seemed that the latter was primary. There was a positive significant correlation between theexpression of CD147 and MTl-MMP protein in gastric carcinoma .The Spearman coefficient was 0.469, P<0.01 .But we did not see any significant correlation between MT1-MMPmRNA and CD147 or MTl-MMP protein (P>0.05). Conclusions: CD147 and MTl-MMP did heavily expressed on the gastric carcinoma tissues. And they have close relationships ,i.e ,the expression of CD147 protein may be an inducer of MTl-MMP, which could help the invasiveness and metastasis in gastric carcinoma. Detecting the combined expression of them may give an objective indication on judging the biological behavior of the gastric carcinoma.
