Objective: To investigate the association between the polymorphism of urotensin II gene and type 2 diabetes mellitus in Kun Ming.Method: We surveyed SNPs in the urotensin II gene in 166 Kun Ming subjects with Type 2 diabetes mellitus who met the criteria of the World Health Organization (WHO) and control cohort consisting of 80 subjects. (1) detect the fasting insulin and in experimental group using radioimmunoassay. ( 2 ) PCR-RFLP (restrict enzyme fragment length polymorphism)was used to examine the SNPs.3)homeostatic model assessment for insulin resistance (H0MA)-IR were also determined.Result: We identified the SNPs with amino acid substitution in the urotensin II gene,designated S89N in control subjects and type 2 diabetic patients. (1) In the type 2 diabetes group, Asn/Asn genotype frequency (9.6%) and Asn allele frequency (29.8%) was higher than in normal controls (6.2% andl8.1%, respectively). (2) In the type 2 diabetes group, the fasting insulin and HOMA-IR level of carriers of the Asn/Asn or Asn/Ser genotype were higher than those of the Ser/Ser genotype.Conclusion: These results suggest that the S89N polymorphism in the urotensin II gene is associated with the type 2 diabetes mellitus in Kun Ming, and what' s more, the S89N polymorphism in the urotensin II gene has related to insulin resistance.
