Objective:To explore the correlation of long-chain fatty acid elongase 6(ELOVL6)gene rs9997926 single nucleotide polymorphism(SNP)and insulin resistance in patients with type 2 diabetes mellitus.Methods:A case-control study method was conducted in the 161 patients(male 86,female 75)who were the first time visit to the endocrine clinic of Yidu Central Hospital from March2013 to December 2013 which comply with the WHO diagnostic criteria,was diagnosed as T2DM and not treated with oral glucose-lowering drugs and 150 healthy individuals with age and sex matched those in the T2DM group in the same physical examination center.The relevant anthropometric indices were measured in two groups of subjects:height,weight,waist circumference,BMI.Also fasting venous blood samples were collected at the morning and the related biochemical parameters were measured:FPG,FIns,TC,TG,and calculate the insulin resistance index(HOMA-IR)to compare the differences of anthropometric and biochemical indexes between the T2DM group and the control group.The venous blood was extracted by automated nucleic acid extraction(ACM)using magnetic beads method to collect DNA of all subject and polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)was used to detect genotypes of rs9997926 in ELOVL6 gene and genomic sequencing.The differences of anthropometric and biochemical markers FPG,FIns,TC,TG and HOMA-IR were compared among T2DM patients with different genotypes.Genotype frequencies of T2DM group and control group was also compared,and logistic regression analysis of ELOVL6 gene rs9997926 single nucleotide polymorphisms and the occurrence of T2DM.Results:(1)The levels of BMI,waist circumference,TC,TG,FPG,FIns and HOMA-IR in T2DM group were significantly higher than those in control group(t=2.678~28.332,P<0.01).(2)Three genotypes(TT,CC and CT)were detected in ELOVL6-rs9997926 gene in both groups,with TT genotype being the most common in the T2DM group,the CC genotype frequency(14.29%)was higher than that in the control group(5.33%),the difference was statistically significant(x~2=7.507,P<0.05).While there was no statistically significant difference between the two groups in regard to allele frequencys(P>0.05).(3)Three genotypes of T2DM group was statistically significant difference in FPG,FIns,homa-ir,TC and TG.Further analysis and comparison show that patients with CC genotype of FPG,FIns,HOMA-IR index was higher than CT genotype patients,with significant difference(F=2.058,3.262,3.127,P<0.05),patients with CC genotype of FIns,HOMA-IR,TG index is higher than TT genotype patients,with significant difference(F=3.216,2.798,3.566,P<0.05)(4)logistic regression analysis showed that there was a significant correlation between the CC genotype and T2DM(ratio OR=1.83,95%confidence interval 1.02-3.23).There was no correlation between C allele and T2DM(ratio OR=1.72,95%confidence interval0.91-2.84).Conclusion:The rs9997926 locus of ELOVL6 gene has single nucleotide polymorphism.CC gene is associated with insulin resistance in T2DM patients and may be a marker of genetic susceptibility to T2DM.
