Objective To investigate the effect of AP polysaccharide about immunization and Antitumor and restraining metastasis on B16-bearing C57BL/6 mice and their mechanism . AP polysaccharide is isolated from the root of a kind of clinical effective antitumor herbs, and it can promote the immunization and enhance the antibiotic effect. Furthermore, the AP polysaccharide has been proved good tumor inhibition effect on EAC, HepA, HepS and P388 in lab, which show it can promote the effect of chemical therapy and reduce its side-effect. Accordingly, to investigate its further function of antitumor on other cancer and its new target of antitumor effect, this experiment mainly observe the lifetime of B16-bearing mice and find out its restraining effect on tumor and metastasis, serum INF— Y , the distributing in tumor-cell cycle and morphologic transformation about apoptosis and immunohistochemical methods and so on to explore the effect of AP polysaccharide on immunoregulation and antitumor.Methods Adopting some researching methods, such as tumor-cell subcutaneous inoculation, given the polysaccharides from vein, ABC—ELISA method, Flow cytometer (FCM), electron microscope and immunohistochemical methods to investigate the effect of AP polysaccharide on immunoregulation and antitumor.Results AP polysaccharide can raise the tumor inhibition rate on B16 CMM. The experiments show that the highest dose group can obviously restrain the tumor with the rate as 48.67% to that of NS group 24.13%. Butits function of prolonging lifetime is limited. We also find that at three different doses AP polysaccharide has promoted the thymus gland factor of tumor-bearing mice and increased INF- Y content in serum. Furthermore it can raise the proportion of S phase in cancer cell cycle but the influence on distributing in tumor-cell cycle and morphologic transformation aren't so obvious , and we have found classical apoptosis corpuscle through electron microscope. The immunohistochemical results show the AP polysaccharide can reduce the positive expression rate of VEGF, FasL, Bcl-2 and increase that of Bax.Conclusion AP polysaccharide has obvious effect of restraining B16 tumor and prolonging the lifetime of CMM-bearing mice, and it also can promote the thymus gland factor of tumor-bearing mice and increase INF- Y content in serum. The AP polysaccharide can induce tumor-cell apoptosis and reduce the positive expression rate of VEGF, FasL, Bcl-2 and increase that of Bax ,which offer good theoretical evidences of its immunopotentiating activities and antitumor effect in clinic.
