The main purpose of this dissertation is to study the structure and properties, antitumor and immunoregulation activities of a polysaccharide from Spirulina platensis (PSP) and its modified product sulfated polysaccharide from Spirulina platensis (SPSP).PSP which was isolated from Spirulina platensis is made of glucose with molecular weight of 410 KD by HPGPC, 94.18% saccharide content by sulfuric acid-phenol reaction, and no UV and IR absorption of sulfate group. SPSP which was synthesized by chlorosulfonic-pyridine method has UV and IR absorption of sulfate group, and the content of sulfate group is 21.22% by the method of barium sulfate turbidity.MTT colorimetric analysis and cell morphological alteration were used to evaluate cytotoxicities of PSP and SPSP against human tumor cells in vitro, the antitumor effects of PSP and SPSP in vivo were determined on the tumor-bearing mice. PSP had no cytotoxicities on Hela and SMMC-7721 tumor cells at the concentration of 12.5~200 μg/mL, but it had cytotoxicities on KB, SGC-7901 and K562 tumor cells at the concentration of 100~200 μg/mL with less than 15% inhibitory ratio. While SPSP at the concentration of 12.5~200 μg/mL had potent cytotoxic activities towards KB, Hela, SGC-7901, SMMC-7721 and K562 tumor cells and higher inhibitory ratio (15~37%) was observed on KB, SGC-7901 and SMMC-7721 tumor cells. PSP at the concentration of 50~100 μg/mL had no effect on KB and SGC-7901 cells morphology, but SPSP at the concentration of 25~100 μg/mL did lead to morphological changes on the two kinds of cells. PSP (25~100 mg/kg) to some extent had inhibitory effects on S180 and Heps in mice, but with no statistic significance compared with model group. While SPSP had remarkable inhibitory effect on S180 (50 mg/kg) and Heps (100 mg/kg) in mice with the inhibitory ratio of 32.7% and 35.5% respectively.
