Tobacco, a traditional Chinese medicine, is first appeared in the ancient Chinese medicinal literature JinYueQuanShu. It has been used to detumescence, detoxification, pesticides in clinical. Now chemical research indicated that the main active constituents are considered to be alkaloid, putrescine, Fatty ethano.In this paper, the solanesol was extracted by supercritical fluid extraction and subsequently isolated by silica gel column chromatography and preparative LC from the leaves of Tobacco (Nicotiana tabacum L), collected in Shandong province, China and the western region of Cuba. Furthermore the sample was analyzed by NP-HPLC and HPLC-MS, and the assay result showed that the content of solanesol was 99%, its purity complied with the quantity request used as the standard substance of Chinese medicine for qualitative and quantitative analysis.A systemic extraction and isolation method was developed and the quantitative analysis methods were set up for the essential oil, nicotine, polysaccharide, amino acids etc. in two samples of the tobacco collected from China and Guba applied with GC-MS, HPLC, UV and amino acid automatic analysis apparatus. The experimental results showed that the contents of solanesol were 1.84%, 1.43%; 9 and 14 chemical components of essential oil were identified; the contents of nicotine were 27.78%, 26.01%; Polysaccharide were 17.21%, 15.01%; the contents of total amino acids were 4.46%, 12.41% respectively in the tobacco leaves. At the same time, the comparative analysis was carried out for two samples of China and Cuba.As far as we known, this is the first report for the comprehensive research of the chemicals in the leaves of tobacco, especially about the extraction and isolation of high purity of solanesol. The experimental result was a useful guide for medicinal research of tobacco and had the practical values for the comprehensive exploitation of the resources of tobacco.
