Healthy adult fresh erythrocytes or cord erythrocytes were used to prepare carrier erythrocytes loaded with MTX lonely or co-encapsulated with VCR together using a modified hypotonic pre-swelling and isotonic resealing technique. The entrapment rate and recovery rate of healthy adult and cord blood MTX-loaded erythrocytes are good. The concentration of MTX is important for efficiency rate of co-encapsulated, but the concentration of VCR is no significant inference to the co-encapsulation. It is the optimal concentration when concentration of MTX is 10 mg/ml and concentration of VCR is 1 mg/ml meanwhile, the two drugs entrapment rate are ( 26.32 ± 1.93 ) %and ( 60.82 ± 5.03 ) % respectively. The total loading parameters of MTX+VCR-loaded erythrocytes are as well as mono-loaded erythrocytes, and co-encapsulated is more efficient in inhibiting the proliferation of K562 cells compared with un-loaded erythrocytes and MTX-loaded erythrocytes. Erythrocytes are practically capable for using as co-encapsulated delivery system of two antineoplastic agents such as MTX and VCR. Cord erythrocytes can be used as drug delivery system and have wide prospect in the clinical application.
