Healthy adult fresh erythrocytes and Kunming mice erythrocytes were used to prepare carrier erythrocytes loaded with Fe3O4 nano-magnetic ferrofluid (MF)lonely or co-encapsulated with anticancer drug vincristine(VCR)together using a modified hypotonic pre-swelling and isotonic resealing technique. Preparation conditions and observation approaches were optimized. We evaluated the carrier erythrocytes through analysis of percentage erythrocyte recovery, osmotic fragility, storage stability and physiological characteristics of erythrocytes. The total loading parameters of MF+VCR-loaded erythrocytes are as well as VCR-loaded erythrocytes, and it is more efficient in inhibiting the proliferation of K562 cells compared with un-loaded erythrocytes and MF-loaded erythrocytes groups. The results can provide treatment pathway for anticancer drug erythrocyte carrier targeting antitumor immunity in the future clinical application, however, the optimization of the breed of MF still remains to be further investigated. Flow cytometry assay was used to research the molecular level effects of MF and VCR on systemic hematogenic immunologic function by means of natural experimental system in vitro. When MF and VCR were used together, the level of CD4 on T cells(38.87±6.37)% was significantly increased (P<0.01),while the mean fluorescence intensity of CD55(68.79±8.98)% was much lower(P<0.05)when MF was used singly. VCR and MF have no harmful effects to hematogenic immunologic function. MF can even elevate the function of leucocytes.
