Study Of The Polymorphism At The Sp1 Binding Site Of COLIα1 Gene Of The Han Nationality And The Weiwuer Nationality

Objective: With peoples, aging, there are more and more patients whowith senile osteoporosis. Osteoporotic fractures are the most seriouscomplication of osteoporosis, its, very easy take happen to those old man.Osteoporotic fractures decrease their living quality in degree. It stillunknown about the pathogenesis of osteoporosis. Evidence from twin andfamily studies suggests that genetic factors play an important role in thepathogenesis of osteoporosis. So we decide to research in such aspects: (1)Study of the polymorphism at the Sp1 binding site of the collagen type Ialpha 1 gene(COLI 1) of the han nationality; (2) Study of thepolymorphism at the Sp1 binding site of the collagen type I alpha 1gene(COLI 1) of the weiwuer nationality; (3) Investigate if there aredifference about the polymorphism at the Sp1 binding site between the hannationality and the weiwuer nationality; (4) And investigate the relationshipbetween the polymorphism and osteoporosis in the han nationality and theweiwuer nationality. Methods: The DNA genomes of vinous blood were extracted fromrandomly people of the han nationality and the weiwuer nationality. We getthe COLI 1 gene by a Polymerase Chain Reaction. Following PCR, theamplification products were digested with the restriction enzyme, and thegenotype were detected by agarose gel electrophoresis. Then the positiveobject would be detected bone mineral density. All the results wereanalyzed with SPSS (11.0) software. RESULTS: There are three genotypes: SS(G/G) ﹑Ss(G/T) ﹑ss(T/T)at the Sp1 binding site of the COLI 1 gene .(1) All of these 156 people,sgenotype is SS in the han nationality,we didn,t find out other genotype; (2)All of these 150 people,s genotype is SS in the weiwuer nationality,wedidn't find out other genotype; (3) We didn,t get a satisfied product whenwe take the method described by Grant et al at first. So we redesigned theprimers and adopted the restriction enzyme Van91I(PflMI). Conclusion: (1) The data indicate that the G-T polymorphism do notexistence or very rare at the Sp1 binding site of COLI 1 gene of the hannationality;(2) The G-T polymorphism do not existence or very rare at theSp1 binding site of COLI 1 gene of the weiwuer nationality; (3) therearen,t difference about the polymorphism at the Sp1 binding site betweenthe han nationality and the weiwuer nationality; (4) we found noassociation between the Sp1 genotypes of the COLI 1 gene andosteoporosis in the han nationality and the weiwuer nationality.
