Investigation On Antimutagenesis Chinese Herbal Medicine Formula And Head And Neck Tumor Prevention

The objective of this experiment is to define Chinese herbal medicine formula to pretect head and neck tumor high-risk group from cancer.Chinese herbal medicine mixture for tumor prevention is based on the theory that there is a close association between mutagen sensibitity and tumor susceptibility.In the theory:many groups,many mutagen sensibitities, the higher mutagen sensibitity ,the higher tumor susceptibility. The individuals with high mutagen sensibitity are found increased chromatosome break in metaphase or increased micronucleus rate in interphase cell when induced by mutagen. Hsu TC suggested heredity instability theory based on mass investigation that the group with instability genome are liable to more tumor susceptibility,and other scholar gradually confirmed the theory. Tumor occurrence need multi-procedure.Activating of protooncogene, inactivation of anti-oncogene and growth factor expression out of control are important link in tumor occurrence.There are close correlation between chromosome break site, fragile site and oncogene site , anti-oncogene and growth factor.Inactivation of anti-oncogene, activation of protooncogene and growth factor expression out of control induced by mutagen lead to tumor occurrence.It is certified that the mutagen sensitivity of head and neck tumor patients is higher than general group. Therefore,high mutagen sensitivity means high tumor susceptibility. The material that could decrease mutagen sensitivity have the quality of stabilizing chromosome and then prevent tumor occurrence. Many reports on chinese herbal medicine of anti-mutagenesis appeared recently.That means many chinese herbal medicines have the quality of anti-mutagenesis,namely the quality of stabilizing chromosome and the protect DNA from mutagen damage. Chinese medicinal herbs could prevent tumor occurence since they could protect DNA from carcinogen.After selecting chinese herbal medicine with quality of anti-mutagenesis,we match them and check their potence on anti-mutagenesis by experiment. It has not appeared in medical periods of investigation on Chinese herbal medicine formula of anti-mutagenesis at present. In the study, L9(34) orthogonal design was adopted. All the Chinese herbal medicines applied in the experiment are confirmed of anti-mutagenesis and not of mutagenesis by the department of medical genetics. The four Chinese herbal medicines –ginseng, slenderstyle acanthopanax root-bark, agaric, glycyrrhizia-is allocated in one formula and the other four Chinese herbal medicines-ginseng, slenderstyle acanthopanax root-bark, tuckahoe, chinese angelica-is in the other formula. Both Chinese herbal medicine formula are used in three levels:zero,average(frequently used dose), high (double of frequently used dose ). The results are that one formula is composed of 20 mg ginseng , 20 mg slenderstyle acanthopanax root-bark, 10 mg tuckahoe, 5 mg chinese angelica and the other formula of 20 mg ginseng, 20 mg slenderstyle acanthopanax root-bark, 10 mg agaric, 5 mg glycyrrhizia. Thereafter,the Chinese herbal medicine formulas are applied in anti-mutagenesis experiments. The participants add up to 150 who are allocated in three groups. There are 50 in the healthy control group who are blood donors and are of physical fitness.There are 50 head and neck tumor patients in the patients group and 50 the first relatives of head and neck tumor patients in the first relative group. The micronucleus test and mutagen sensibitity experiment with BLM as mutagen are performed. The results manifest:there is no striking difference of autonomic b/c value among the three groups(p>0.05). A notable difference of autonomic micronucleus rate was seen between the groups of the first degree relative group, the patients group and the the healthy control group(p<0.01).
