My experiment aim toestablish culture process of (NSCs) from embryonic rat brain,it can be a basic for cell transplantation in the future, and wecan observe the effects of glutamate and nimodipine on the(NSCs).The method is (NSCs) culture in vitro. We can obtain (NSCs)from embryonic rat mesencephalon tissue, culture in (NSCs) medium,obtain a lot of neurospheres. At the same time, we have only oneneurosphere continue to culture in differentiation medium, we can seea lot of cells from neurosphere migrated out, they can increase inamount with time, the cells from the neurosphere have not onlyneuron but also neural glial in morphology. Identified the neurosphereand the cells from the neurosphere in immunocytochemistry. They arenestin, GFAP and NSE positive respective. Above all, there are theability of self-renewing and multipotential differentiation in theculture, they conform to (NSCs). In short, we obtain the model of(NSCs) culture in vitro.In this foundation, we added glutamate into the (NSCs) medium,observe the effect on the (NSCs). It has been more reported thatglutamate have toxic (injury) effect on (NSCs). In this experiment,added glutamate into (NSCs) medium in different concentration, MTTmethod evaluate the effect of glutamate on the (NSCs).It has been reported that there have more drugs object to theinjury effect of glutamate on the neural cells. But there is less reportedabout calcium ion antagonist (nimodipine) have protective effect onglutamate caused neural cell injury. In the course of experiment, weadded different concentration nimodipine, we use MTT method toevaluate the protective effect on glutamate caused injury of (NSCs).The results of experiment show that glutamate can reduce thesurvival rate of (NSCs).
