Objective To observe the alteration of serum cTnT, TNF-α , MDA contents and MPO activities in myocardium, and to investigate the protection of Puerarin (Pur) from the early myocardium damage of severe scalded rates. Methods A total of 88 Wistar rats were randomly divided into three groups: the control (n=8), the recovery (n=40) and the treatment (n=40), and in the later two groups a scald burn comprising 30 percent of the total body surface area full-thickness were established. The activity of serum CK-MB and the content of cTnT and TNF- α were detected in 1, 3, 6, 12 and 24 hours after the burn, as well as the content of MDA and the activity of MPO in myocardium . The morphologic changes were observed by electrical microscope. The indexes above were analyzed to acknowledge the variations in three groups with SPSS software. Results (1) In the recovery group cTnT, TNF- α and MDA, MPO went up 1 hours after burns, while CK-MB went up 3 hours after burns, and all attaining the top at 12 hours post burn. In cTnT, increasing about 20 times of the control group at 12 hours post burn. (2) In the treatment group, the indexs above had the same trends as the recovery group. But comparing with it, cTnT were much lower at 1, 3, 6, 12, 24 hours, TNF- α and CK-MB were much lower at 3, 6, 12, 24 hours, MPO and MDA were much lower at 1, 3, 6, 12 hours. (3) A series of ultrastractual changes in the myocardium were found at 3 and 6 hours postbum, the above changes attained the worst in 24 hours. Comparing with the recovery group, the pathological changes were obviously alleviated in the treatment group. Conclusions (1)The cardiadc myocytes damage occurred in early stage of seriously scald.
