Viral diseases have high incidence and high prevalence, which are dangerous to human's health, and now they are thought of more and more, especially after the discovery of AIDS and its pathogen virus. Varicella-zoster virus (VZV) belongs to Herpesviridae and human is its only host. It is the pathogen of varicella and herpes zoster. In immunocompromised individuals, especially the HIV infected, severe complications may be accompanied with. Neuralgia is characteristic in herpes zoster, and in older and weak patients it can be severe and intolerant, and go on for a long time, even when the lesions have disappeared. So the effective prompt antiviral treatment is important to extenuate complications and neuralgia. In recent years, the appearance and development of new antiviral medications provide more choices to control VZV infection, e.g. acyclovir, ganciclovir, and so on.Objective: This experiment is based on Vero cells culture to isolate wild-type VZVs, and then to evaluate the cytotoxicity to Vero cells and the antiviral activity to the wild-type VZV and the standard strain VZV-Ellen of ganciclovir and acyclovir in vitro.Methods: ①To isolate and identify wild-type VZVs by Vero cells culture. ②The isolated and identified wild-type VZV4 and the standard strain VZV-Ellen were titrated, and the median tissue culture infective dose (TCID50) were calculated.③The sensitivity of Vero, Hela and McCoy to VZV were detected. ④The cytotoxicity of ganciclovir and acyclovir to Vero cells were detected, and the median toxic concentration (TC50) and the maximal nontoxic concentration (TCO) of ganciclovir and acyclovir were detected. ⑤Antiviral activity of ganciclovir and acyclovir to the wild-type VZV4 and the standard strain...
