Objective: ①To detect the content of mesaconitine(MA) in 6% tinctura aconiti by high performance liquid chromatography (HPLC) method.②To compare the percutaneous speed(J) of MA with different pH buffer solution.③ To compare the accelerative effect of three physical factors on percutaneous absorption of aconite alkaloid: ion-introduced therapy of the direct current, pulse direct current and phonopbresis therapy. ④To observe the percutaneous speed(J) when the corneum is moved.Methods: We crashed R.aconniti into powder to extracting the alkaloids, and obtained 4g/ml aconiti extract. Use the extract as delivery solution. To detect the color spectrum graph of MA and HA in the extract. Adult rats were randomly divided into four teams including seven groups,5 rats each. Each group made separated skin for percutaneous absorption. The samples were obtained from receiving solution after treatment of 10min, 20min, 30min, 40min, 50min,separately. A high performance liquid chromatography ( HPLC) method was established to analysis the MA graphs of the samples. All data collected were analyzed with SPSS 11.5 software. Peak area(A) — concentration(C) standard curves were obtained. Calculate Q (accumulativepermeation quantity) at each correspond time. We obtain the J (percutaneous speed) from the Q-t curve and compared them in each team.Results: CD The content of mesaconitine was 0.0319mg/ml in 6% tinctura aconitini.(2)With the buffer solution of pH4.0,the J of MA in free percutaneous absorption group is 4.068 iig/cm2 ■ min, and in another direct current-accelerated group for MA ion-introduced ,J is 12.623 i£g/cm2 ? min. There is obvious statistics difference, (P<0.05). ?With the same direct current intensity, when the pH of buffer solution were 4.CK 7.CK 9.0, J is 12.623 iig/cm2 ■ mim 4.916 iig/cm2 ? mi^ 3.121 iig/cm2 ? min, respectively. There are obvious statistics differences between any two groups (P<0.05).@ With the buffer solutions of pH4.0,J of phonopbresis therapy is 6.11l4g/cm2 ■ min. J of direct current group is 12.623 4g/cm2 ? min and pulse direct current is 12.268 ng/cnT ■ min. There is not statistics difference between the post-two groups(P>0.05).obvious statistics differences exist both between the first group and the second group(P<0.05),and between the first group and the third group(P<0.05). (5) J of normal rat skin is 4.168 ijg/cm2 ? min. J of the corneum-moved group is 39.993 iig/cm2 . min. The latter is 9.60 times of the former. There is obvious statistics difference between two groups (P<0.05). 6) Within 50 minutes .the accumulative permeation quantity of mesaconitine increases in all groups.Conclusion:1 .The content of mesaconitine is about 0.053 % in 6% tinctura aconiti.2. With the buffer solution of pH4.0, the direct current has obvious effect in aconiti percutaneous absorption.3.In the same conditions, when the pH of buffer solution decreases, the percutaneous absorption of MA increases.4.In the experiment of aconiti percutaneous absorption in vitro, we findthe direct current and pulse direct current both have obvious effect on accelerating the transdermal absorption of mesaconitine. Percutaneous speed of direct current is 3.03 times of free absorption; percutaneous speed of pulse direct current is 2.94 times of free absorption. The phonopbresis therapy has not statistic ffect.5.In the same conditions, the percutaneous speed of corneum-movedskin is about 9.60 times of that of normal skin.6. Within 50 minutes, the accumulative permeation quantity of mesaconitine increases under all conditions.
