Objective: To explore the effect of inhale glucocortioids on the nuclear factor-KB expression and the degree of cytokines in BALF of murines with asthma to analysis the relationship between them.Methods: 30 BALB/c mice were divided randomly into normal control groups asthmatic groups and inhale Pulmicort Respules( budesonide) group.(1) The asthmatic groups were primed with intraperitoneal injection by 1 0ug OVA and 20 mg AL (OH ) 3 on the frist day,the seventh day and the fourteenth day.On days 15-18, the mice received two consecutive airway allergen challenges of 2 % OVA solution delivered by aerosolization to set up murine model of atopic asthma.(2)The treatment group were primed with intraperitoneal injection by 10ug OVA and 20 mg AL (OH) 3 on the days of 0,7,14. On days15-18, the mice received two consecutive airway allergen challenges of 2%OVA solution delivered by aerosolization.But before inhale 2%OVA, the mice inhale Pulmicort Respules eath time.(3)The normal control groups were primed and challenged by 0.9 % NaCL solution.Within 24 hours after the last challenge,the broncho-alveolar lavage fluid(BALF)and the lung of the mice were collected. The expression of nuclear factor-KB ( NF-kB ) were observed by immunocytochmical staining; the levels of interleukin-4( IL-4) interleukin-5(IL-5 ) interferon-γ(IFN-γ) in BALF were measured by ELISA.The total cells and eosinophils(Eos)of the BALF were counted.Results: l.The total WBC (11.73 + 5.18) and EOS counts (8.0+3.87) in asthmatic group were significantly higher than those of control group (5.29+2.61-.0.25 + 0.35) and budesonide group (7.25 + 3.9, 0.8 + 1.01) (P<0.01)2.The level of IL-4 (95.23 + 22.82) and IL-5 (90.97 + 20.27) in BALF of asthmatic group were significantly higher than those of control group (36.83+ 11.15, 38.01 ±13.50) and were significant reduced by budesonide. (45.58+14.89, 49.2 + 24.68) (P<0.01)3.The percentage of cells of active NF-kB of asthmatic group (25.6+ 5.76) were significantly higher than those of control group (8.6 + 3.98) and were significant reduced by budesonide (12.10 + 2.56) (P<0.01)4.There were good positive correlation between the percentage of cells of active NF-kB and the degree of IL-4, IL-5 level in BALF (r=.0.95, 0.92, t=16.1, 12.4. P < 0.01)Conclusion: l.The level of IL-4, IL-5 in BALF of the asthmanal mice are significantly higher than those of control group which suggest that the Th2-type immune responses occupy a dominant position than that of Thl .2.Nuclear factor-KB of the asthmanal murine is over-activated.3.The activation of NF-kB has positive correlation with the level of IL-4, IL-5 in the BALF ,which suggest that NF-kB may play an important role in the regulation of the expression of many interleukins in asthma.4. Inhale glucocortioid can reduce the level of IL-4, IL-5 in the BALF and over-activation of NF-kB can be reduced by inhale glucocortioid.lt suspect that glucocortioid may be restrain the function of Th2 cells throw which the Thl-type immune responses is outstanded.
